چکیده

Several candidate-genes for Rheumatoid Arthritis are studied using multi-marker association scores based on 839 cases and 855 unrelated controls from the North American Rheumatoid Arthritis Consortium (NARAC) as part of the Genetic Analysis Workshop 15 data. Strong signals previously identified on PTPN22 and PADI4 were repeated in our findings. We also found significant association evidence that leads to an association network among the loci of PTPN22, PADI4, DLG5, SLC22A4, SUMO4 and CARD15. To account for false positives, we used permutations to have the family type I error rate controlled under 1%.

Case-control association analysis of rheumatoid arthritis with candidate genes using related cases

We performed a case-control association analysis of rheumatoid arthritis (RA) for several candidate genes using the North American Rheumatoid Arthritis Consortium (NARAC) data provided in Genetic Analysis Workshop 15. We conducted the case-control association analysis using all related cases and unrelated controls and compared the results with those from the analysis of samples using only one r...

Genetic association with rheumatoid arthritis-Genetic Analysis Workshop 15: summary of contributions from Group 2.

The papers in presentation group 2 of Genetic Analysis Workshop 15 (GAW15) conducted association analyses of rheumatoid arthritis data. The analyses were carried out primarily in the data provided by the North American Rheumatoid Arthritis Consortium (NARAC). One group conducted analyses in the data provided by the Canadian Rheumatoid Arthritis Genetics Study (CRAGS). Analysis strategies includ...

Current understanding of the genetic aetiology of rheumatoid arthritis and likely future developments.

Most of the work described herein was performed by the North American Rheumatoid Arthritis Consortium (NARAC). After a brief description of the NARAC and the multiplex family resource that has been developed by this consortium, we will summarize the current status of genome-wide screens using this valuable family collection. Next, we describe work that is under way to further delineate the gene...

Replication of putative candidate-gene associations with rheumatoid arthritis in >4,000 samples from North America and Sweden: association of susceptibility with PTPN22, CTLA4, and PADI4.

Candidate-gene association studies in rheumatoid arthritis (RA) have lead to encouraging yet apparently inconsistent results. One explanation for the inconsistency is insufficient power to detect modest effects in the context of a low prior probability of a true effect. To overcome this limitation, we selected alleles with an increased probability of a disease association, on the basis of a rev...

Application of bivariate mixed counting process models to genetic analysis of rheumatoid arthritis severity

We sought to i) identify putative genetic determinants of the severity of rheumatoid arthritis in the NARAC (North American Rheumatoid Arthritis Consortium) data, ii) assess whether known candidate genes for disease status are also associated with disease severity in those affected, and iii) determine whether heterogeneity among the severity phenotypes can be explained by genetic and/or host fa...
